Epithalon (Epithalon (Epitalon / Epithalone))

Category: Anti-Aging. Status: Research compound.

A synthetic tetrapeptide based on pineal gland peptide epithalamin. One of few compounds shown to activate telomerase in human somatic cells.

How it works

Activates telomerase reverse transcriptase (hTERT) expression for telomere maintenance. Also normalizes circadian rhythm via melatonin regulation and modulates antioxidant enzymes.

Key facts

  • Molecular weight: 390.4 Da
  • Half-life: 3-4 hours
  • Bioavailability: High (SubQ)
  • Storage: Lyophilized: -20°C. Reconstituted: 2-8°C for 14 days.

Dosing overview

  • Typical dose: 5-10 mg
  • Frequency: Daily for 10-20 days
  • Duration: 10-20 day courses, 2-3x per year
  • Route: Subcutaneous injection

Protocol notes

  • Run as short intensive courses, not continuously: 5-10 mg daily for 10-20 days.
  • Repeat the course 2-3 times per year rather than dosing year-round.

Reported benefits

  • Telomerase activation
  • Telomere lengthening
  • Melatonin normalization
  • Improved sleep patterns
  • Antioxidant enzyme upregulation
  • Potential lifespan extension

Possible side effects

  • Injection site reaction
  • Drowsiness
  • Altered sleep timing (initially)

Research

  • Peptide regulation of telomerase in human somatic cells (2020): Activated telomerase in fibroblasts, extending proliferative potential by 10 additional doublings.
  • Epithalon effects on melatonin and aging markers (2019): Restored nocturnal melatonin peak in elderly and improved antioxidant status.

  • recovery_midwest rated it 5/5 - steadier sleep, but my dreams got weird: i ran epithalon for about 12 weeks while juggling grad work on the east coast and trying to keep up with training after 50, and it really seemed to smooth out my sleep and morning grogginess. the weird part was my dreams got intense and kind of annoying, but for me the tradeoff was still worth it.
  • vik_r rated it 1/5 - Sleep stayed weird, and that was the dealbreaker: The one thing I wanted most was steadier sleep, since I write on the East Coast and my schedule gets messy fast, but after about 3 weeks on Epithalon I did not get that. I kept waking up too early, and a few nights I felt kind of wired for no clear reason, which was annoying because I was trying to protect recovery and keep my days sharper. It could have been coincidence, but for me it was enough to stop. I did not notice any clear upside that made the sleep weirdness worth it, so I came away pretty disappointed.
  • mom_honest rated it 4/5 - helped my 3am wakeups, but the weird dreams were real: i tried epithalon after a stretch of 3am wakeups had me feeling like a zombie on clinic days, and yeah, i was curious about the anti-aging hype too, imo. after about 3 weeks, i started sleeping more straight through, and that was the main thing i noticed. i also had a couple nights of really vivid dreams, kinda weird but not a dealbreaker. could've been placebo, but for me it was enough to make me keep it in mind.
  • protocol_harley rated it 5/5 - Skeptical at first, but my sleep really shifted: I went in pretty sceptical, ngl. I used Epithalon for about 4 weeks because teaching leaves me oddly wired at night, even here in Sweden when I want a proper reset. For me it seemed to smooth my sleep and morning feel enough that my doubt actually changed.
  • early_rows rated it 5/5 - Sleep finally settled down for me: 5 stars. For me, the big change was sleep, it got properly steady after about 10 weeks on Epithalon. I’m in Ireland, working sales and training around that, so broken nights were doing my head in. Could be just my own experience, but I felt a lot more rested.
  • dana_n rated it 4/5 - Subtle sleep shift over three weeks: I tried Epithalon because my sleep had gone a bit patchy after a heavy work stretch, and I wanted to see if it helped my recovery while I was training in Singapore heat. In the first week, nothing dramatic. By week two, I was falling asleep a little easier and waking less often. Week three felt steadier still, with slightly better mornings. For me, the change was gentle, not dramatic, and the only caveat was that it took patience.
