N-Acetyl Semax Amidate (N-Acetyl Semax Amidate (NA Semax))
A more stable, longer-acting version of Semax with acetyl and amidate modifications that resist enzymatic breakdown. Used intranasally as a nootropic for focus, BDNF support, and neuroprotection.
How it works
A heptapeptide analog of ACTH(4-10) that increases BDNF and NGF expression, modulates the dopaminergic and serotonergic systems, and provides neuroprotective and pro-focus effects. The acetyl/amidate modifications extend its half-life versus plain Semax.
Key facts
- Molecular weight: ~900 Da
- Half-life: Longer than base Semax (modifications resist peptidases)
- Bioavailability: Intranasal (primary); subcutaneous also used
- Storage: Refrigerate (2-8°C); keep nasal solutions cold and use within the labeled window.
Dosing overview
- Typical dose: ~300-600 mcg per day
- Frequency: 1-2x daily
- Duration: Cycled (e.g. weeks on/off)
- Route: Intranasal spray/drops
Protocol notes
- Common nootropic use is a few hundred mcg intranasally, 1-2x daily.
- Often dosed in the morning/early afternoon to avoid overstimulation at night.
- Frequently cycled (e.g. a few weeks on, then a break) and stacked with Selank.
Reported benefits
- Improved focus and mental clarity
- Raises BDNF/NGF (neurotrophic)
- Neuroprotective
- Longer-acting than base Semax
Possible side effects
- Nasal irritation
- Overstimulation/insomnia if dosed late
- Mild headache (uncommon)
Research
- Semax and BDNF expression (2016): Semax increased BDNF and related neurotrophic signaling in the hippocampus in animal models.
