Anyone cycling rapamycin for longevity and tracking sleep or labs?

I’ve been reading up on rapamycin and keep coming back to the same practical question, which is how people are actually timing it in real life. The idea of pulsed dosing makes sense to me on paper, but I’m still trying to work out what’s worth watching beyond the theory.

Has anyone here noticed anything useful with sleep, energy, skin, or recovery after a few months on it? I’m especially keen on what changed, if anything, when you compared before and after against basic markers like lipids, fasting glucose, or just how you felt in the arvo.

I’ve personally only done the reading so far, not taken it yet. The mouth sores and the wound healing piece make me a bit cautious, because that seems like the sort of thing you would only want to discover after you’ve committed to a dumb haircut or a gym injury. From what I’ve seen, low-dose weekly use is the usual discussion, but I’d rather hear how it has played out for people in practice, good on ya if you’ve tracked it properly.

  • mateo_h: i’ve been on a low‑dose weekly schedule for about 4 months now, usually 1 mg on monday, then off for six days. i started tracking sleep with a ring and doing a basic lipid panel every 6 weeks. for me the sleep numbers are pretty much unchanged, maybe a couple minutes deeper on the first few nights after a dose but it settled. energy in the afternoon feels a bit steadier, but i can’t say it isn’t just better pacing my workouts. my fasting glucose stayed in the low‑80s the whole time, lipids were
  • formernico: Sounds like a solid start, mate. For what it’s worth, I’ve also seen the sleep dip flatten out after the first week and my HRV stayed flat too. The steadier afternoon energy lines up with what I felt when I timed my runs – maybe just a subtle mTOR tweak. Have you tried shifting the day‑off window to see if the early‑night deep sleep bump moves?
  • dana_n: Anecdotally I saw a tiny dip in my REM after the first dose too, but it normalised after a week. My fasting glucose has hovered in the low‑80s as well, and triglycerides dropped modestly – I think diet played a bigger role. The steadier afternoon energy felt real for me, though I also tightened my training schedule.
  • humblequinn: The REM dip line up with what I was worried about – I was hoping it’d settle quickly, so it’s reassuring it normalised after a week for you. I’m curious, did you notice any change in skin texture or wound healing when you tightened your training? I’m still thinking about a weekly 5 mg schedule and will probably start with a single dose to see how my sleep reacts before adjusting.
  • humblequinn: That’s exactly what I was wondering about – I’ve been pencilling in a Sunday off and taking the dose on Mondays. I’ll give your idea a go and move the rest day to Thursday, then see if the deep‑sleep bump slides later in the night. If I notice any shift in my morning HRV or the afternoon slump eases, I’ll update. Thanks for the tip, mate.
  • humblequinn: Thanks for the rundown, mateo_h. The 1 mg Monday dose lines up with what I was thinking, so that’s useful. I’m curious – did you do any blood work right after a dose to see if there’s a short‑term dip in triglycerides or anything, or just the 6‑week panels? Also, have you noticed any impact on wound healing or mouth sores when you push a hard leg day?
