Pulsed Rapamycin, Sleep, Energy and Skin: my first week

I started a once‑weekly 4 mg pulse of rapamycin a couple of weeks ago because I read that it can trigger autophagy and help with ageing. I’ve had no major issues so far, but I noticed that my sleep has become a bit lighter – I fall asleep quicker but wake up more often in the middle of the night. Energy during the day is steadier, with fewer crashes after my training sessions, but I do get a mild metallic taste in my mouth on the days after the dose.
The skin feels a little tighter and I’ve seen a subtle reduction in fine lines compared to before I started. I’m a bit worried about the potential for mouth sores and the reported lipidaemic rise, though I haven’t seen any change in my cholesterol yet. Anyone else with a similar routine?

- miles_codes: I’m in the same boat. I’m doing a 4 mg pulse every week and the taste and light‑sleep are still there after a month. For me the taste fades after the first day or two and I never got a sore. The skin feels firmer and lines look a bit smoother, but I’m waiting for a lipid panel to see if there’s any rise. Do you notice any change in your cholesterol or any sores? I’ve kept my caffeine low in the evenings, which seems to help with the nighttime awakenings.
- hank_m: Tbh i haven’t done a lipid panel yet but i didWeekly check the chart and i still see normal total cholesterol so far, no spikes, no aches, and the taste lingers a day or so after the dose then fades just like yours, the light sleep is still a thing but caffeine cuts it down a bit.
- liam_sleepnerd: I’ve checked my panel a couple of months after starting, and the LDL and triglycerides were unchanged – still in the mid‑70s for LDL. I haven’t had any injection‑site soreness yet, just the usual mild itch that fades after a day. The caffeine cut hasn’t made a huge difference for me; I still wake a few times but it’s less insomnia‑like. What’s your caffeine window?
- greg208: I’m only three weeks in, so the panel still looks like the baseline – LDL 72, TG 140, nothing up so far and still no mouth sores. , even if I only have a cup in the afternoon – I’ve found that pushing it earlier cuts the odd mid‑night wake but it’s still a bit of a “stitch” if you’ll forgive the metaphor. The metallic taste hasn’t cleared, but it fades after the dose day. Goma.

- greg208: Thanks mate, I’ve kept my evening coffee to a minimum too. I haven’t seen any obvious change in my cholesterol yet – the last check was just a week ago and it was fine. No mouth sores so far, but I do still get that metallic taste for a couple of days after the dose. After the first week my sleep was lighter, and it’s slightly improved but still wakes a few times.
