Rapamycin: how did it affect my immune response after a marathon?

Posted by recovery116 in Longevity & Anti-Aging - 5 points, 6 comments.

I started rapamycin 3 mg once a week after my last 100k. I read that low dose pulsed rapamycin can boost immune function and mimic caloric restriction. Fwiw i had been getting a flu shot a month before the race.

After the run i didn’t feel super sick, but i did notice less soreness and a faster bounce back the next day. I did notice a tiny mouth sore, but it resolved in a few days. I’m not saying rapamycin cured anything, just that the recovery felt a touch smoother than the last time i ran with just nutrition and sleep.

Since rapamycin also activates autophagy, i think that might explain the cleaner muscle feel. I’m still tracking my energy, sleep, and any side effects. If anyone else has tried this protocol or has data on how rapamycin affects post‑race recovery, let me know.

  • hank_m: Tbh i’ve been on a 3 mg weekly pulsed rapamycin routine since a few 100‑k runs and i’ve seen some similar vibes to yours – less soreness, quicker bounce‑back, and a slightly smoother muscle feel that i suspect is the autophagy bit kicking in. I did get a tiny mouth ulcer early on, but that cleared up in a couple of days just like you described. I বিরুদ্ধে keep protein up, stay hydrated, and i’ve been noting my sleep quarters and heart rate changes; the only side‑effect i’ve really felt is a mild
  • mom_sean: Yeah, the itch came on after the first dose but it faded fast. I haven’t done any cytokine tracking – that’s a lab thing. I focus on sleep, protein and staying hydrated. Still, the smoother recovery has stuck with me after each 100k. Fwiw, it’s all anecdotal.
  • aspiring_trailrun: That’s a good sign you’re on the right track. I haven’t pinned down cytokines either, but after the first couple of weeks I logged resting HR and sleep stages – HR fell by a few beats and I’d get more deep‑sleep time. The itch you mentioned is common and usually gone by 24–48 hrs. Any shift you’ve seen in inflammatory markers?
  • recovery116: Thanks for the update, aspiring_trailrun. I did notice a drop of about 4‑5 bpm in resting HR the day after the run and an extra 20‑30 min of deep sleep overall, so it lines up with what you saw. I didn’t do any cytokine panels, but the muscle soreness was lighter and the bounce back was quicker – the mouth sore cleared in about 48 hrs like you said. Fwiw, i plan to get a basic CBC and CRP next week to see if there tempting changes there. Have you noticed any shift in those markers?
  • recovery116: I saw the itch too, after my first dose. It faded quick, same as your experience. I still had that tiny mouth sore but it went away in a few days. Do you notice any change in muscle soreness after the 100k with rapamycin? Also, what hydration routine works best for you? 🙌
  • recovery116: Fwiw i did notice the same mouth sore and a quick itch early on, so maybe that’s a common idiosyncratic reaction. I haven’t tracked cytokines yet; thinking about a simple panel after the next run. Do you keep an eye on HRV or any blood markers? Would love to compare numbers if you have them. 🙌
