First week on 3 mg rapamycin weekly – what to expect?

I just hit the one‑week mark on 3 mg rapamycin once a week. I’m 38, a software engineer, and this is my first pill for longevity. The pharmacist told me that the drug’s half‑life is about 62 hours, so it should stay in my system for a while.

I’ve been taking it after dinner, with a 500 mg dose of metformin in the morning to keep my glycogen high. The first day I woke up with a slight sore mouth, which I think is normal at low doses. No severe GI upset so far, but I’ve been noticing a mild improvement in my skin tightness, maybe the autophagy is kicking in.

I’m still monitoring my lipids because I read rapamycin can raise them. I’m keeping a log and plan to check my fasting glucose next week. Any experience with rapamycin + metformin combos?

  • sarah_c: I’m a bit surprised you’re seeing skin tightness after just a week, but that could just be a mild placebo effect or natural skin changes – for sure not a proven rapamycin sign. I’ve tried rapamycin with metformin too, keeping doses low, and I did see a slight dip in fasting glucose but my lipids did trend a touch higher after a month, so I started a higher‑dose omega‑3 and started watching the LDL. Have you checked your cholesterol yet? And how’s the appetite? I’ve felt a little hungry lately.
  • hank_m: Tbh i just got my cholesterol panel last week, the LDL was 4.6 mmol, a tad up the way you mentioned, so i splashed on 2 g omega‑3 and added a handful of avocados to my lunch, and i’ve noticed my appetite isn’t as gnarly as it was earlier. I’ll let you know how it holds up in a couple of weeks.
