My First Month Trying Low‐Dose Weekly Rapamycin – What I’ve Noticed

I started a 4‑week trial of 4 mg rapamycin once a week, the dose that most longevity clinics prescribe for anti‑aging. Not gonna lie, the first week I woke up with a sore mouth and a bit of nausea, probably the GI upset that shows up on the side‑effect list. After that, I noticed my sleep a little deeper – I fall asleep faster and feel less groggy in the morning.
My body feels lighter, and I’ve had fewer infections over the past month, which is weird but cool. The only real downside has been a mild increase in cholesterol, so I’m keeping an eye on my lipid panel. I’ve been on a stable 1000 mg metformin daily, so this is the first new pill in a while.
I’m not saying this is the whole secret to longevity, but for me, the combination of rapamycin with metformin feels like a nice balance between boosting autophagy and keeping my blood sugar in check. Any thoughts from folks who’ve been on this regimen longer?

- trevor_recovery: I’ve read up on weekly rapamycin a lot, but I never actually tried it. My own experiment with metformin (500 mg twice a day) was mainly for blood‑sugar control; I didn’t notice a big change in sleep or infection rate. The GI upset you mentioned lines up with the studies, but the cholesterol rise is a red flag that I also saw in a few reports. I’m curious, how often do you re‑check your lipid panel, and have you noticed any other metabolic shifts that might need tweaking? Also, have you tried pa
- brandon245: I’ve been pulling my lipid panel every six weeks now, so the jump in LDL/HDL got me a little nervous. No other big changes, glucose still in the low‑normal range, blood pressure steady. I’ve kept rapamycin alone so far; I’m not on any peptides yet, just the metformin. Will look into AKP‑I once I see the numbers settle. Any other folks doing that combo?
- brandon245: I’m checking my lipid panel every two months now, start of each 2‑month cycle. Apart from the cholesterol bump, I haven’t seen big swings in glucose or insulin; my fasting numbers stay in the same range. I haven’t paired rapamycin with AKP‑I yet, I’m just running it solo and watching the numbers.
