Spermidine alongside weekly rapamycin, redundant or complementary for autophagy

Been running rapamycin weekly for a few months now and starting to wonder about stacking spermidine on top. Both hit autophagy but through different pathways right, rapamycin via mTOR and spermidine through polyamine signalling and acetyltransferase inhibition. Seems like they could complement each other but for what it is worth i cant find much on whether combining them actually adds anything or if its just paying twice for the same effect.

I eat heaps of aged cheese and wheat germ already so probably getting some dietary spermidine anyway. The supplement doses seem to range from like 1mg up to 10mg daily which is a pretty wide spread. Anyone here actually run both together and noticed anything different versus rapamycin alone? Energy, recovery, skin, anything measurable?

Also keen to know if people have tracked any biomarkers while on spermidine solo. The long term human data still seems pretty thin from what i can tell. Just trying to figure out if its worth adding or if im better off letting the rapamycin do its thing on its own.

  • sleep_coastal: I’ve been on weekly rapamycin for about six months and added a low‑dose spermidine capsule (around 2 mg) most mornings for the last eight weeks. For me the combo didn’t feel like a big jump in energy or recovery compared to rapamycin alone, but I did notice a subtle uptick in morning alertness that seemed a bit smoother, maybe less of the occasional fog I get on rapamycin days. Skin looked a touch more hydrated, though that could just be the extra cheese I’m eating. I haven’t tracked any labs y

The morning alertness thing is interesting, i get a bit of brain fog on rapa days too so if spermidine smooths that out even slightly thats worth noting. 2mg daily seems like a sensible entry point given im probably already getting some from diet. Are you planning to keep the spermidine dose at 2mg or titrate up? And when you say a full cycle before bloods, what timeframe are you thinking, like another 3-4 months?
