Question about rapamycin and autophagy cycles

ive been reading up on mTOR inhibitors and rapamycin /peptides/rapamycin seems to be the big one for longevity. coming from a chemistry background i get the basic maths of inhibiting mTORC1 to trigger autophagy, but i reckon the timing is where it gets tricky. not gonna lie, i am a bit nervous about the immunosuppression side of things or getting mouth sores. i have seen some folks mention pulsed dosing once a week to keep things safe, but i am still new to this whole biohacking scene.

do any of you lot actually track biomarkers to see if it is working or just go by how you feel? i am keen to optimise my cellular recycling without messing up my immune system. i was also looking at spermidine /peptides/spermidine as a milder way to get that same clean up effect since its naturally in things like aged cheese. would combining the two be overkill or is that a common way to do it? just my own curiosity here, so if anyone has experience with the cycle timing, i would be chuffed if you could share.

  • sofia_h: I’ve Anchorage tried a once‑a‑week pulse of 1‑2 mg rapamycin on a 48‑hour off window, mostly to keep mTOR down without dragging my immune line. I keep an eye on a full blood count and C‑reactive protein every 3 months – if the white cells dip or CRP spikes I pause. I’ve also taken 2 mg spermidine nightly; it feels less jaw‑tightening than rapa and I’ve not noticed extra ‘sore mouth’. For me the combo seemed fine, but I’d still compare the two in one month’s worth of tests before going longer.
  • sarah401: Thanks for the detail, Sofia. I’ve also been doing a weekly 1.5 mg pulse and checking my full blood count every two months – haven’t seen any drop in neutrophils yet. I’m curious, have you noticed any difference in sleep quality when you add the spermidine, or does it feel neutral?
  • greg208: 5 mg pulse and fortnightly FBCs sound solid – not gonna lie, I’m still juggling whether to start with a 2 mg pulse or roll it in a month. I haven’t noticed any change in sleep since adding spermidine, still feels neutral to me. Do you ever see any mouth sores or other skin side‑effects with that dose? And how do you nab the neutrophil data, routine NHS sample or a private kit?
  • greg208: great advice, thanks. 2 mg/L – and I’ll start 1 mg once a week with a 48‑hour break as you do. I’m curious: how long after the first month did you spot any shift in your CRP or WBC? Also, did the “no sore mouth” stay consistent or did it pop up after a few weeks?
