Curious if anyone has tried PE 22-28 or P21 for mood/cognition

Hey all. I've been digging into some of the less-talked-about cognitive peptides and wanted to see if anyone has hands-on impressions of a couple of them.

First one is PE 22-28. The TREK-1 angle is what caught my attention. I like the idea of something that works on a different pathway than the usual SSRI stuff, and the preclinical neurogenesis data is interesting to me on paper. I have zero human experience with it though, and the safety profile is basically a blank page. Just wondering if anyone has even messed with it for mood or general cognitive feel.

Second is P21. The CNTF-derived piece that skips the appetite suppression sounds clever, and the fact it crosses the BBB is a plus in my book. Again, I haven't pulled the trigger. The limited human data makes me want to hear from someone who has actually tried it.

For me personally, I've had decent responses to the Semax family (Adamax included) for focus days, but I'm always curious about what else is out there that hits neurogenesis without the rough edges.

Anyone here run either of these? Pure anecdote territory for me, just curious what real users have noticed.

  • tracker_jade: I’ve only messed with PE 22-28 once, just a low dose for a week to see if it lifted my mood at all. Anecdotally I felt a slight edge of calm in the afternoons, but it was so subtle I could’ve been imagining it, no clear boost in focus or memory. I didn’t notice any side effects, but the lack of human data makes me hesitant to go longer. Has anyone tried stacking it with something like Semax for a clearer effect? For P21 I haven’t tried it yet, just waiting on more real‑world reports.
  • humble_dad: The afternoon calm thing is worth noting. That tracks with the TREK-1 mechanism, right? Potassium channel modulation could plausibly take the edge off without hitting serotonin directly. I don't think I'd stack it with Semax though, just because I'd want to isolate what each one is doing first. Running two novel compounds together makes it impossible to attribute effects. Maybe save stacking for after I get a feel for PE 22-28 solo for a few weeks.

The TREK-1 mechanism makes sense to me on paper but I keep coming back to the same question, what does it actually do in a human brain and what are the downsides. Preclinical neurogenesis is great but rats aren't us. Stacking is where my curiosity goes too. If you did try it, pairing with something that hits BDNF pathways seems like the obvious play. But again, guessing. I'd rather hear from one person who actually ran it solo for a few weeks before I even
