Old review on GHRPs and how MK-0677 came from GHRP-6, still useful context

Saw this older ScienceDirect review on growth hormone-releasing peptides and their analogs. It walks through GHRPs in animals and humans, and traces how the non-peptidyl secretagogues like L-692,429 and MK-0677 were designed using GHRP-6 as a starting template. Good background read if you want to see where the whole GHRP family really started.

For me the interesting part is how MK-677 is basically a pill version of the same idea GHRP-6 was working on decades ago, just tweaked to last longer in the body. The review makes it clear these were developed as legitimate research tools, not gym hacks, which I think gets lost sometimes in forums.

Anyone here still running a GHRP-6 or ipamorelin based stack alongside CJC, and noticing the oral MK-677 research ever since? Curious if the older mechanism still holds up in practice.

The origin story from GHRP-6 to the small molecule mimics is genuinely one of the cleaner examples of rational drug design, and most people here probably haven't seen it laid out that way 👀 To your question, i did a GHRP-6 + CJC stack for a few months last year, felt like the hunger was the only real effect for me, sleep got a bit better but nothing dramatic. Switched to ipa + CJC after and that seemed cleaner for me, less sides. Never
  • tyler589: Haha yeah the hunger on GHRP-6 is no joke, that's the classic give away it's doing its thing via ghrelin receptors. Ipa was cleaner for me too once I swapped, way less munchies and the sleep side felt a bit smoother. MK-0677 in the papers always looked promising for IGF-1 bumps without the injection hassle but the appetite and water retention stuff from what others have said kept me on the pin route tbh...
  • grinder265: Yeah the ghrelin mimic piece is what kills it for a lot of people. The hunger side effect is the whole mechanism honestly, you can't really separate the GH release from the appetite signaling since it's the same receptor doing both. Did the ipa + CJC give you any water retention or was it pretty clean compared to the GHRP-6?
  • tyler589: Yeah the ghrelin receptor thing is the catch right, you can't have one without the other... I haven't done straight GHRP-6 in a while tbh, the hunger was too much even at low doses. For me ipa + CJC was cleaner on water, like noticeably less puffiness in the face and hands within the first week or two. GHRP-6 used to make me hold a bit of water, especially if I dosed close to bedtime. Could just be me but the ipa side seemed more dry overall. Are you running both together or stacking something
