VIP peptide caught my eye, anyone looked at the CIRS/mold research?

I've been digging into VIP lately, partly because I keep hearing it mentioned in the chronic inflammatory response stuff that comes up around mold illness and CIRS, and partly because the mechanism actually makes sense to me. From what I've read it works through VPAC1 and VPAC2 receptors and basically calms down inflammatory cytokine production while also supporting vasodilation and T reg activity, which is a pretty broad claim but at least it's a real pathway and not just hand waving. The half life is absurdly short though, like a minute or two in plasma, which makes me wonder how much actually sticks around long enough to do anything meaningful with a nasal spray, and that part bugs me a little because some people talk about it like it's a daily fix.
I haven't tried it myself yet, still in the reading stage, but I'm curious if anyone here has actual experience using it for chronic inflammation or immune stuff, and whether the short half life ends up mattering in practice or if it just means you dose more often. Also is anyone tracking newer human trials on it beyond the Shoemaker-era work?

From what i've seen in the research, VIP seems to work more like a signalling nudge than a sustained drug, which would explain why the nasal route needs repeat dosing rather than a once daily hit. Whether that translates into meaningful changes in someone with chronic inflammation is another matter entirely, and i don't think the shoemaker cohort work is the strongest evidence to lean on at this point. I haven't used it myself for that reason s
- sofia_h: Reckon youre spot on about the shoemaker stuff, that cohort work always felt a bit thin to me too. The signalling nudge idea makes biological sense though, VPAC receptor activity doesnt need sustained exposure to shift cytokine profiles imo. But yeah without proper controlled trials its hard to separate real effect from the enthusiasm around it.

But the signalling nudge idea makes sense to me, it's not meant to sit around in plasma doing work, it's more like a quick "hey calm down" to the immune system. The repeat nasal dosing is a pain but if that's how it actually works then fair enough. Would love to see proper controlled trials too but reckon we'll be waiting a while on that one.

My only remaining question is whether the repeat dosing thing actually hits that threshold often enough, like if you're spraying twice a day is that frequent enough to keep the signal going or is it just peaks and valleys the rest of the time?
