Trying low‑dose rapamycin – anyone else doing weekly pulses?

Hey all, tbh I’m pretty new to the whole longevity stack and I’ve been reading about rapamycin as the “most studied” anti‑aging drug. The page says a typical low‑dose is 3‑6 mg once weekly, half‑life around 62 hours, and that it can boost autophagy and even improve vaccine response. I decided to give it a quiet try last month – started with 3 mg on a Saturday night after a light dinner, then skipped the next week to see how I felt.

For me the only thing I noticed was a slight sore spot on the inside of my cheek a day or two after the dose, which cleared up on its own. Sleep seemed a tad deeper, but maybe that was just the extra rest I took that weekend. I’m curious if anyone else has stuck with the weekly schedule, tried a different day of the week, or adjusted the dose up to 5 mg?

Also any tips on monitoring lipids or other labs while on it would be appreciated 🧬. Thanks!

  • aspiring_codes: I’ve been on a 4 mg weekly rapamycin pulse for about 4 months now (usually Saturday morning with coffee). For me the biggest change was a subtle shift in morning energy – not a huge boost but I felt a bit more “clear‑headed” after the first few weeks. My cheek didn’t react, but I did get a mild dry mouth the night after dosing, which cleared by morning. I’ve kept an eye on lipids by doing a basic lipid panel every 6 weeks. My LDL nudged up a couple points, so I added a low‑dose omega‑3 and made
  • gina_zone2: Thanks for the detail, aspiring_codes – the Saturday morning coffee timing sounds doable, I’ll probably try that instead of night. The dry mouth thing is good to know; I’ll keep a water bottle handy just in case. I’m planning a lipid check in about 5 weeks, so I’ll follow your 6‑week interval and maybe add a fish oil capsule if LDL creeps up. Did you notice any change in appetite or cravings after your pulses?
  • amber464: Interesting, I’ve also seen a slight dry‑mouth thing after my first dose, so that lines up. Do you track fasting glucose at all? I’ve been curious if that subtle “clear‑headed” feeling ever coincides with any changes in my morning glucose trends.
  • gina_zone2: tbh I haven’t been checking fasting glucose yet, but your dry‑mouth note makes me think I should start a simple log – maybe note morning numbers before and after each 3 mg dose to see if any trend shows up. Good call, thanks 🧬
