DOI as a 5-HT2A research tool, why does it get ignored in longevity talk?

Saw a paper pop up recently using DOI to map 5-HT2A receptor distribution in aged mouse tissue, and it got me thinking. Most longevity and biohacking chatter skips right over the serotonergic side, but 5-HT2A signaling ties into neuroplasticity, sleep architecture, and probably some of the mood/cognition stuff people chase with other compounds.

The DOI page here on ResearchSafe calls it the standard pharmacology tool for this receptor, and the half-life is long enough that the data must be pretty clean compared to shorter-acting agonists. What caught my eye is the vasoconstriction and BP angle. If 5-HT2A is something we actually care about as we age, the cardiovascular hit is a real caveat and not something to handwave.

I am curious whether anyone here thinks 5-HT2A is a meaningful target for cognitive aging, or if it is mostly a dead end outside of psychedelic therapy research. And for those who follow this space, do you see longevity researchers picking it up more, or staying away?

  • patient_codes: I’ve played around with low‑dose psilocybin a few times just to see how it felt on focus and sleep, and I did notice a bit smoother winding down at night, but also a slight tightness in my chest the next morning that made me wonder about blood pressure effects. It’s hard to tell if that’s just anxiety or something vascular, and I’ve never tracked BP tightly enough to say for sure. I think the receptor is interesting for plasticity, but the cardiovascular caveat keeps me cautious about pushing it
  • theo_benchpr: Yeah the chest tightness thing is exactly the kind of signal that made me flag the BP angle in the first place. Hard to know if it's anxiety versus vascular without numbers. I have not tracked HRV around psilocybin specifically, but I do wear a watch that records it overnight. Next time I dose I'll check the night-of and next-morning readings against a baseline week. Even a rough before/after comparison might tell us something. I'll report back if I get a clean window.
  • grinder265: That chest tightness thing is exactly why I keep eyeballing the 5-HT2A vascular angle before touching anything in this space. 5-HT2A agonism is known to drive vascular smooth muscle contraction, its not subtle in the pharmacology literature. Have you ever tried tracking HRV with a chest strap the morning after, or were you just going by feel? Feel alone is kinda unreliable for separating anxiety from actual vascular tone changes, ngl I'd wanna see resting HR and BP both before calling it one or
  • theo_benchpr: Honestly I never strapped a monitor on, just went by how my chest felt and a cheap cuff reading when I remembered. Reading your reply I realize that's pretty lazy data on my end. Going to start logging resting HR and BP the morning after, and maybe grab a chest strap to track HRV overnight to see if there's actually a signal or if I'm just pattern-matching anxiety onto normal soreness.
