New fisetin senolytic trial out, intermittent dosing still looks smarter than daily

Saw a fresh paper land this week building on the Mayo fisetin work, this time looking at intermittent "hit" dosing versus daily in older adults. The hit-and-run group kept the senolytic signal and had fewer GI complaints, which honestly matches how I have been timing mine around heavy training blocks for a while now. Daily just feels like you are asking for tolerance and gut issues for no extra benefit.

The interesting part is they are finally tracking actual functional outcomes, not just biomarker stuff. Walking speed, grip strength, that kind of thing. Still small numbers and short follow up, but it is a step in the right direction.

What I keep wondering is whether the absorption problem is the real ceiling here. Most fisetin studies use formulations that boost bioavailability, and I do not see anyone talking about that gap in the popular write ups.

Anyone here pair it with a fat source or a liposomal version and notice a difference on how you feel the next day? Curious if that is placebo or actually doing something.

  • patient_codes: I've been doing the same thing honestly, just dosing it around my hardest training days and skipping the rest of the week. For me the next day I feel less of that general beat up feeling in my joints, but it could totally be placebo, I never did a strict on off comparison. The absorption point is the one that bugs me too. I tried a liposomal version a few months back and honestly couldn't tell if it was doing more than the regular capsules I had been using. Maybe the difference is real and just
  • weekendnate: Yeah, the joint soreness thing matches what I see too, less of that "creaky" feeling 24 hours after a hard session. Could easily be placebo for both of us, honestly, but it's the kind of signal I'd rather have than not. On the liposomal question, I had the same problem telling them apart. I started tracking morning HRV on both and the spread was basically noise, like 3 to 5 ms either way depending on sleep. So I went back to regular powder mixed into yogurt or whatever fat I had around, and the
  • rachel_ketogen: Honestly the liposomal question is one i keep going back and forth on too. Theres some pharmacokinetic data showing higher peaks but idk if that translates to anything functional for senolytics since the whole theory is you dont need sustained exposure. My approach lately is just regular caps, timed a couple days before my hardest lifts, and i watch HRV the next morning. If its flat i assume i did too much, fisetin or not. The joint feel thing you mentioned is interesting though, i might actual
  • weekendnate: Yeah the pharmacokinetic versus functional gap is what bugs me about the liposomal pitch too. Higher blood levels sound great on paper but senolytics are supposed to work in a hit, so chasing a flatter curve seems backwards to me. Liking the HRV check the next morning, that is a clean way to keep yourself honest. I will steal that framing for my next block. For me the joint feel thing is subtle, maybe one or two workouts out of five where my knees feel noticeably less crunchy, but I am not gonn
