Beta-defensin peptides, the immune thing I keep reading about, anyone looked in?

Came across a profile on ResearchSafe about beta-defensin peptides and honestly the framing was new to me. I always thought of these as just "natural antibiotics" the body makes on skin and in lungs, but the writeup talks about them also calling in dendritic cells and bridging innate and adaptive immunity, which is a much bigger claim than I expected for something so small.

What caught my eye is the wound healing and biofilm disruption angle. I get mild dermatitis flare ups in this weather and I am way more interested in a peptide that helps the skin barrier talk to immune cells than in slathering on another cream. The half life being only a few hours is annoying for anything practical though, would mean frequent dosing or some kind of delivery trick.

Is anyone here actually tracking beta-defensin research beyond the obvious skin and mouthwash studies? Curious if it is going anywhere real or if it is one of those things that looks great in a dish and then never makes it past phase 1. Also has anyone seen human dosing data, because the 50 to 200 mcg range on the profile is pretty wide and I would love to know what people are actually working with in trials.

  • elle187: The half life thing is what kills it for me too, a few hours means you are basically dosing all day or hoping some delivery system actually works in humans and not just in a petri dish. I had not looked at beta-defensins since nursing school honestly, back then it was more the antimicrobial angle. The dendritic cell recruiting piece is newer to me and sounds promising on paper but i would want to see actual human data before getting excited, since a lot of immune modulating stuff looks amazing
  • elle_z: Honestly I didn't mention a dose range in my post so not sure where you're getting the 50 to 200 mcg from, maybe you saw it on the same profile page? The one I was reading was just a general ResearchSafe entry, didn't have concrete dosing that I recall.   • fasted_scientist: Yeah the in vitro to in vivo drop off is brutal for immune stuff, half the time the concentration needed in a dish is wildly unrealistic compared to what you could ever get in tissue. HBD-2 is the one most papers point to for the dendritic cell recruiting claim but yeah I have not seen anything beyond phase 1 for skin indications if memory serves.
