Spermidine log week 3, plus a question about stacking with rapamycin later

Okay so I started spermidine about three weeks back, the polyamine kind, and I genuinely did not expect to feel anything but my face looks less puffy in the mornings and my skin just looks a bit more even, could absolutely be placebo or could be the autophagy thing finally kicking in, I am not going to pretend I can tell. I am taking it once a day in the morning on an empty stomach and that seems to work fine, no GI stuff for me so far which I know can happen. The thing that got me curious enough to start was reading about it inducing autophagy and basically mimicking caloric restriction without the actual fasting, and at 30-something in Singapore with the kind of sleep and training load I put myself through, anything that helps cellular clean-up sounds worth a shot to me.

Now the question part, has anyone here tried layering spermidine with low dose pulsed rapamycin down the line? I know rapamycin also hits the autophagy pathway through a different mechanism (mTOR inhibition) and I am trying to think through whether stacking makes sense or if that is just asking for too much immunosuppression. Would love to hear what others running a longevity stack have done.

On the rapamycin question, that is a whole different beast mate, you are talking about a prescription immunosuppressant that needs a doc on board and proper bloodwork, not something you just slot in next to your spermidine and hope for the best. The question i would push back on is whether stacking them even makes mechanistic sense in the first place though, since both seem to hit autophagy pathways, you would want to know if you are just overstimulating one route while leaving others idle.
  • sanjay_j602: Honestly yeah that is the exact thing that was nagging me too, like if both are pushing the same mTOR suppression pathway are you just doubling up on one lane while the lysosomal stuff sits there doing nothing. Curious if anyone with more biochem background can actually speak to whether pulsing rapa at like 5mg once a week even does anything meaningful on top of spermidine, or whether it is basically just expensive redundancy πŸ˜…
