Anyone tried layering Pal-GHK with rapamycin for skin + cellular side?

New to all this so bear with me. I have been reading up on the Matrixyl family and rapamycin separately, and I am curious if anyone here is stacking them in a way that makes sense.

For skin, Pal-Tripeptide-1 seems like the basic building block since it signals fibroblasts to make new collagen. Pal-Tetrapeptide-7 is the one that calms IL-6 and stops the breakdown side. So in my head they cover different parts of the problem, build plus protect.

Then rapamycin at low pulsed doses for the cellular longevity piece, autophagy and all that. Could just be wishful thinking but the idea of addressing skin aging from the topical side while also supporting cellular cleanup systemically feels like it would cover more ground than either alone.

For anyone who has run something like this, how did you time it? Did you keep rapamycin weekly and do the peptides daily, or space things out differently? And did you notice anything on skin texture or recovery, or is it too early to tell? Just trying to learn from people who have been at it longer than me.

  • fasted_scientist: Bit of a mismatch here, ngl. Pal-Tripeptide-1 and Pal-Tetrapeptide-7 do different jobs on paper, sure, but the evidence for either doing much topically is pretty thin. A few in vitro studies showing fibroblast activity, hardly anything properly controlled in real skin at real world concentrations. Then pairing that with rapamycin is where it gets daft, sorry. Rapamycin is a serious drug with a real side effect profile, even pulsed. Using it as a stack partner for a face cream ingredient that mi
  • dad217: Fair points, and I appreciate the honesty. The penetration thing is something I had not really thought about, I just assumed if it was in the formula it was doing something. You are right that rapamycin is not a casual add-on. I have not started anything yet, just trying to learn what people who actually do this have learned the hard way. When you say bloodwork, what would you actually want to see before even considering a longevity stack like this?
  • mike65: Their reply got cut off but yeah, the rapamycin part is what stood out to me too. I keep seeing people online talk about it like it's some harmless longevity vitamin and it's not. Real drug with real sides, anecdotally people on it deal with mouth sores and lipid stuff. I'd want actual doctor supervision before going near it, not just a podcast guy saying it's safe.
  • dad217: Yeah rapamycin is the part that gave me pause too. I am not touching it without bloodwork and a doc who actually knows the dosing, because I keep reading the same thing you said, mouth sores and lipid shifts are not rare. Was just thinking out loud about the theory of pairing them. Appreciate you pushing back on the "longevity vitamin" framing, that is kind of exactly why I asked.
