Klotho keeps coming up in my longevity reading, has anyone actually tried anything for it?

Ok so i've been going down a bit of a rabbit hole on klotho after seeing it mentioned alongside more common longevity stuff like rapamycin and NMN. From what i understand it's a protein we make less of as we age, and higher levels seem to track with better cognition and slower aging in the research, but the supplements angle is still pretty murky to me. I know there's no real established human protocol for it and the peptide/protein version is not something you can just pop like a capsule.
I'm mainly curious if anyone here has been tracking it in bloodwork, or found a way to nudge levels through lifestyle stuff like fasting, sleep, or other compounds they were already taking. I did see some preclinical stuff about it being neuroprotective but i don't want to over-read mouse data. For context i'm in my mid 30s, mostly focused on cognitive upkeep and general healthy aging, and i'm already on a pretty standard stack with metformin and weekly low-dose rapa. Would love to hear what people smarter than me are doing or reading on this one, the klotho entry on this site is a good starting point but i'd like more practical takes.

- hank_v: Honestly i've been poking around the same rabbit hole lately, mostly from the cognitive upkeep side since that seems to be where the human data is most interesting (the klotho gene variant studies on cognition are actually quite robust, those are worth a look). I'm not tracking it in bloodwork though, from what i understand the standard tests measure klotho in blood but whether that actually reflects brain klotho is genuinely debated. So it might not tell you much even if you paid for it. On t
- elle_z: Yeah the blood klotho vs brain klotho thing was something i kept bumping into too, that's partly why i never bothered paying for the test. Sounds like we're reading similar stuff lol. Exercise keeps popping up as the boring reliable answer huh, i've been decent about zone 2 stuff but honestly my lifting has been on and off. The klotho variant studies are what got me hooked, such a weird finding that one variant seems to track with cognition so strongly. Have you changed anything in your routine
- coastal_biohacker: Yeah the blood vs brain klotho question is the part that tripped me up too. If serum levels don't really track with what's happening up there, then paying for the test feels kinda like flying blind. Exercise being the cleanest signal makes sense. Seems like one of those things where the boring answer (move your body) is the best one we actually have ✨
- elle_z: Haha right?? The "boring answer wins again" thing is so real for longevity stuff. Honestly exercise was already in my routine but knowing there's an actual mechanism for klotho on top of everything else makes me want to stop skipping my zone 2 days 😅 did you notice anything subjectively from training more, or is it more of a "trust the biomarker" move for you?
