Anyone tried IDRA-21 alongside a Semax/Selank routine?

I've been running Semax in the morning and Selank later in the day for a few weeks now, mostly for work focus and to keep my mood steady when training gets heavy. The combo feels solid but I'm curious if there's room to push the cognitive side a bit further without going overboard on stimulation. I keep reading about IDRA-21 and the idea that a single dose can keep working for days because of how it sits on AMPA receptors.
That long tail is appealing tbh, less stuff going in and out of my system every day. My main worry is excitotoxicity, since Semax already has my glutamate system humming along. Would stacking IDRA-21 on top be asking for a headache or worse, or do they actually play nice together?
If you've run something similar I'd love to hear how you timed it and whether the focus felt clean or just wired. Also how often are you actually dosing it, every few days? Trying to figure out if this is worth exploring or if I should just stay where I am imo...

- sarah401: Honestly I tried something in this ballpark last autumn, semax in the am and idra-21 a couple of times a week spaced apart from the semax dose... For me the first week felt like genuinely clean focus, almost too clean, then by day 5 I started getting a low grade tension headache that I could not shake off. Could just be my system being sensitive to ampakoids, but my hrv also dipped a bit on those days which I did not love. What made me drop it was exactly the excitotoxicity question you raised.
- lifter_robin: That's a really useful data point actually, the HRV dip is the bit I'd watch too. I haven't tried IDRA-21 myself but I've been wary of ampakines for similar reasons, the long half life sounds convenient until you're stuck with side effects you can't wash out quickly. Did the headache and HRV bounce back within a few days of stopping, or did it linger? That'd tell you a lot about whether it was acute stimulation or something more cumulative...
- omar_v: Yeah that tension headache thing tracks, ampakoids can do that. The HRV dip is a good signal to actually pay attention to too, ngl. I'd probably keep semax and selank as the base and skip adding IDRA unless something else feels like it's missing. Have you tried tweaking the semax schedule at all? 👀
- cycles_dad: Omg yes the HRV drop is what made me pump the brakes honestly... I've been doing Semax like 2x a week instead of daily lately and it seems smoother. Do you run yours on training days only or just spread it out?
- cycles_dad: Oh that's a and honestly I didn't track it as carefully as I should have, just kinda felt off for almost a week after stopping. The headache was gone in a day or two but my sleep and HRV took longer to normalize, maybe 5 or 6 days before I felt like myself again. That kinda freaked me out tbh, which is why I haven't gone back to it. The long tail thing is great in theory until you have a bad reaction and you're just stuck riding it out... Imo that's the part that doesn't get talked about enough
- cycles_dad: Yeah that "too clean" feeling is exactly what I'm a little scared of honestly, like when focus gets so sharp it almost feels unnatural... The HRV dip is a good data point too, I track sleep scores but haven't paid much attention to HRV, gonna start logging it if I try this. Curious how far apart you spaced the semax and idra doses, like morning vs same day evening or a full day off between them?
- weekendnate: Tbh I started on training days only, like 4x a week, but found my focus tanked on off days when work got gnarly so I spread it out to a small dose daily and bumped up on heavy sessions. Your HRV drop makes me want to pull back and try Mon/Wed/Fri/Sat only and see what my morning readings do. Got a Whoop or anything tracking yours?
