Trying PE 22-28 for a week – any mood changes noticed?

I decided to give PE 22-28 a go after reading about its TREK-1 blocking action and the preclinical mood data. I used a very low dose, just a couple of micrograms intranasal each morning, for seven days straight. I kept a simple notes file on my phone, writing down how I felt each evening and rating my mood on a 1-5 scale.

For me the first two days felt pretty neutral, maybe a tiny lift in energy but nothing strong. On day three I noticed I was a bit less reactive to small stressors, like the usual morning traffic annoyance seemed to fade quicker. By day five I had a sense of calm that lasted into the evening, though I also had a mild headache one afternoon that went away after drinking water and resting.

I cannot say if this was the peptide, placebo, or just a quieter week at work. I stopped after seven days because I want to see how I feel off it before considering another round. If anyone has tried it, I would be curious to hear your timeline and any side effects you noticed. Remember to check with a doctor before trying anything new.

  • tiredmira: I tried PE 22-28 a few months back at a similar low dose intranasal each morning. For me the first three days felt pretty flat, just a slight bump in focus that could have been placebo. Around day four I noticed I didn’t dwell on little frustrations as much – the usual email ping that would annoy me seemed to roll off easier. By day six I had a steadier calm that lingered into the night, though I did get a mild pressure headache one afternoon that went away after water and a short break. I stop
  • leah_s: It’s interesting that we both noticed the calmer reaction around day four and a mild afternoon headache that eased with water. Did you track any sleep changes during your week?
  • weekendnate: Sounds familiar. I ran PE 22-28 intranasal low dose for five days and noticed the same delayed calm around day four, though I didn’t get a headache. My mood drifted back to normal two days after stopping, which felt like a clean washout.
  • leah_s: It is interesting that you also felt that delay. For me day three was the shift, but day four or five is when it really felt stable. I am glad you did not get the headache, maybe I just was not drinking enough water. I am curious if your mood drift back was sudden or just gradual?
